抗红鳍东方鲀盾纤毛虫药物的筛选及组织分布研究

摘要:
为筛选出抗红鳍东方鲀盾纤毛虫的天然化合物,并评价其对红鳍东方鲀的安全性,选取了二十种中药有效成分,在光学显微镜下观察统计药物各浓度梯度在10 min、30 min和60 min时对盾纤毛虫的杀灭作用;选择杀虫效果最好的两种药物进行杀虫率检测,通过棋盘法进行联合用药评价;并使用高效液相色谱法研究两种药物在红鳍东方鲀体内的组织分布。结果表明:MNL、NHK两种化合物与盾纤毛虫作用10 min时,药物浓度为8 μg/mL可观察到杀虫作用,药物浓度达到32 μg/mL时杀虫作用显著,随着时间延长杀虫作用增加,但二者联用呈拮抗作用;MNL和NHK在红鳍东方鲀肌肉中达峰时间分别为0.75 h和1 h,在肝脏均呈现双峰现象且第一达峰时间为0.5 h,并分别于3 h和12 h在受检组织中消除。研究表明,即此两种天然化合物具有显著的杀虫作用,在一定范围内具有剂量依赖性和时间依赖性,药物吸收快、达峰时间短、消除快,应用中宜单独用药不宜联用。

Screening and tissue distribution study of natural compounds against scuticociliatid ciliates from Takifugu rubripes

Abstract:
In order to screen out natural compounds against scuticociliatid ciliates and evaluate the safety to T. rubripes, twenty ingredients of traditional Chinese medicine were selected. We observed and evaluated the killing effect of the drugs on ciliates at 10 min, 30 min and 60 min under the microscope. Two compounds with the best insecticidal effect were selected to detect the insecticidal rates and evaluate the combination index through checkerboard techniques, and HPLC were used to study the tissue distributions of the two compounds in T. rubripes. The results showed that when MNL and NHK acted on ciliates for 10 min, the insecticidal effect was observed at a drug concentrations of 8 μg/mL, and the insecticidal effect was significant when the concentrations reached to 32 μg/mL, but the combination of the two showed an antagonistic effect. The peak times of MNL and NHK in the muscles were 0.75 h and 1 h, and both showed double peaks in the liver and the first peak time was 0.5 h. Besides, the two were eliminated in the tissues at 3 h and 12 h, respectively. Studies indicated that the two natural compounds have significant insecticidal effects, dose-dependent and time-dependent within a certain range, and possess fast drug absorption, short peak time, and fast elimination. In application, it is advisable to use drugs alone rather than combined.
